Abstract
This seminar will explore the critical steps for a smooth flow of T-cell engineering. The talk will introduce the concepts of chimeric antigen receptor (CAR) or T cell receptor (TCR) non-viral CRISPR/Cas-mediated knock-in in the T cell genome. In addition, we will focus on technical aspects of the production of homology-directed repair templates (HDRT), T cell activation, T cell electroporation-regeneration and expansion, quality control by flow cytometry, potency assays and in vivo testing. Further, we will discuss further developments in the field.
