GUIDE-seq™

Nomination assay

SeQure DX's GUIDE-seq is a cell-based, orthogonal nomination assay that validates and reinforces ONE-seq™ findings to support regulatory requirements. GUIDE-seq utilizes co-transfection of a DNA tag alongside your gene editing tool of interest to profile potential off-target effects in your chosen cell type.

ONE-seq for Variant-Aware Therapeutic Guide Selection
Shown here in a line graph are cumulative counts of candidate off-target loci based on their ONE-seq scores. All loci with assigned ONE-seq scores are represented here. As ONE-seq score increases, number of off-target loci decreases dramatically, indicating that majority of sites do not show clear evidence for cleavage in the ONE-seq assay.
